Abstract

Weissella cibaria CMU(oraCMU)´Â ±¸°­ °Ç°­À» ÁõÁø½ÃÅ°´Â ÇÁ·Î¹ÙÀÌ¿Àƽ½º Áß ÇϳªÀÌ´Ù. Æ÷¸£ÇǷθ𳪽º ±ä±â¹ß¸®½º¸¦ ºñ·ÔÇÑ ´Ù¾çÇÑ Ä¡ÁÖ º´¿ø±ÕÀº Ä¡ÁÖÁ¶Á÷ Æı«¸¦ À¯¹ßÇÏ°í Ä¡ÁÖÁúȯÀ» À¯¹ßÇÑ´Ù. ÀÌ·¯ÇÑ °á°ú´Â ¿­ºÒÈ°¼ºÈ­ oraCMU°¡ ÀáÀçÀûÀ¸·Î Ä¡ÁÖ¿° ¿¹¹æ¿¡ È¿°úÀûÀÎ ÇÁ·Î¹ÙÀÌ¿Àƽ½ºÀÏ ¼ö ÀÖÀ½À» ½Ã»çÇÑ´Ù.

Weissella cibaria CMU (oraCMU) is reported as one of the probiotics that promote oral health. Various periodontal pathogens, including Porphyromonas gingivalis (P. gingivalis), can cause periodontal tissue destruction and periodontal disease. P. gingivalis infection in human gingival fibroblasts (HGFs) induces the expression of proinflammatory cytokines and enhances the production of matrix metalloproteinases (MMPs). The present study investigated the inhibitory effects of heat-inactivated oraCMU on the mRNA expression of proinflammatory cytokines and MMPs in P. gingivalis LPS-stimulated HGFs. Cell viability was performed using the MTS assay. The mRNA expression of proinflammatory cytokines, such as interleukin-1¥â (IL-1¥â) and, IL-6, tumor necrosis factor-¥á (TNF-¥á), and MMPs, such as MMP-1 and MMP-3, was examined by reverse transcription-quantitative PCR (RT-qPCR). Heat-inactivated oraCMU dose-dependently inhibited the mRNA expression of IL-1¥â, IL-6, and TNF-¥á in P. gingivalis LPS-stimulated HGFs. Also, the mRNA expression of MMP-1 and MMP-3 dose-dependently decreased. These results suggest that heat-inactivated oraCMU could be an effective probiotic for preventing periodontitis.
